Assay of angiogenesis

Pro- and anti-angiogenic effects

The angiogenic processes in HDMEC-model is very sensitive to effects of drugs. Pro-angiogenic drugs increases the density of capillary-like structures, while anti-angiogenic drugs suppresses the formation of angiogenic sprouts. Drug effects can be analysed by software supported images analysis (SSIQ).

Human endothelial cells are cultured under different conditions. For analysis, the cells are visualized by fluorescence staining of the cytoplasma.

Control

Cultured HDMEC are stimulated to form capillary like structures.

Pro-angiogenic drugs

Hypoxie Autokon

Addition of a pro-angiogenic drug increases the formation of capillary network.

Anti-angiogenic drugs

Addition of anti-angiogenic drugs prevent the stimulation of angiogenesis and the formation of capillary-like structures is clearly reduced.

The angiogenic process is documented by image analysis, yielding detailed quantification (e.g. dose-response curve etc.). We demonstrated the effect of several drugs in HDMEC system.
