Assay of angiogenesis
Angiogenesis in three dimensional capillary-like network
Human dermal microvascular endothelial cells (HDMEC) can be stimulated by specific effectors to develop capillary-like structures. This process is visualized by fluorescense (Calcein-AM, Hoechst 33342) staining.
Cross-section through an angiogenic sprout. The three dimensional capillaries show tube-like structure with a central Lumen (L).
ECM: extracellular matrix
D: debris
EC: endothelial cell
The three dimensional HDMEC system allows:
- screening of potential inhibitors or activators of angiogenetic processes
- quantification of anti-angiogenic drug effects on the development of capillary-like structures
- quantification of anti-angiogenic drug effects on migration, proliferation, cell-cell interaction and apoptosis/necrosis
- analysis of mechanisms behind the detected drug effect
